My lab studies congenital heart disease and heart failure in mouse models and human. Our results have led us to develop major interest in the role of insulin resistance and metabolism in both diseases. First, we have discovered that maternal age is an independent risk factor in a mouse model of congenital heart disease. The maternal age effect is related to a factor in the mother (and not the oocyte) that affects cardiac development. The data strongly suggest that the risk is related to a reversible abnormality likely related to insulin resistance or obesity. Second, we are studying the mechanism of secondary insulin resistance in a mouse model of dilated cardiomyopathy. A clinical research project that builds upon this work indicates that there is a significant incidence of secondary insulin resistance in pediatric heart failure patients as well.
